Clinical studies of semaglutide generally measure glycated hemoglobin, fasting plasma glucose, body weight, and composite cardiovascular endpoints. The SUSTAIN program enrolled adults with type 2 diabetes, while the STEP program focused on obesity without diabetes. Administration follows a stepwise escalation schedule designed to limit gastrointestinal effects during the first weeks. Reported outcomes include mean percentage weight change, the proportion of participants reaching defined weight-loss thresholds, and rates of nausea, vomiting, and diarrhea. Long-term data on durability after treatment stops are still limited and remain a topic of ongoing research.
Semaglutide is a synthetic peptide analog of human glucagon-like peptide-1, developed by Novo Nordisk and first approved in 2017 for type 2 diabetes. It belongs to the incretin mimetic class, a group of agents that reproduce the glucose-dependent actions of endogenous GLP-1. The molecule was engineered to resist degradation by dipeptidyl peptidase-4 and to bind serum albumin, extending its half-life from minutes to roughly one week. Approval for chronic weight management followed in 2021, based on large cardiovascular and obesity outcome trials.

Semaglutide is a synthetic peptide that acts as an agonist at the glucagon-like peptide-1 receptor. It is a structural analogue of human GLP-1(7-37), modified to resist enzymatic degradation by dipeptidyl peptidase-4. The peptide backbone contains alpha-aminoisobutyric acid at position 8, a substitution that stabilises the helix and slows cleavage. A fatty diacid side chain attached through a linker at lysine 34 promotes binding to serum albumin, which extends the circulating half-life. These two modifications together allow less frequent administration than native GLP-1 requires.
Activation of the GLP-1 receptor couples to Gs signalling and raises intracellular cyclic AMP in pancreatic beta cells. The resulting insulin release depends on prevailing glucose concentrations, so the effect is greater when glucose is elevated. Receptor engagement also suppresses glucagon secretion and slows gastric emptying, which flattens post-meal glucose excursions. In the central nervous system, signalling in hypothalamic and brainstem regions is associated with reduced appetite and lower energy intake. Studies continue to examine effects on cardiac, renal and hepatic endpoints; whether those benefits are independent of weight change remains an open question.
Solid peptide material is generally kept at reduced temperature to limit degradation. Short-term storage at 2 to 8 degrees Celsius is common, while longer archival storage at minus 20 degrees Celsius or below is typical for lyophilised powder. Vials should remain sealed and protected from light, because ultraviolet exposure can oxidise susceptible residues. Repeated freeze-thaw cycles are avoided, as they promote aggregation and loss of soluble material. Solutions are less stable than solids and are usually prepared close to the time of use.
Reversed-phase high-performance liquid chromatography is widely used to assess purity and to separate the parent peptide from related substances. Mass spectrometry confirms identity and can resolve modifications that differ by a few daltons. Size-exclusion chromatography detects dimers and higher aggregates, which are relevant to both stability and immunogenicity questions. Peptide mapping with enzymatic digestion locates specific modifications along the sequence. Circular dichroism provides a secondary-structure profile, although it gives limited information about local conformational changes.
Quality control for peptide material focuses on identity, purity, content and the profile of impurities. Common degradants include deamidated and oxidised forms, plus aggregates formed during storage or handling. Forced degradation studies under heat, light, acid and peroxide help define which conditions accelerate change and which analytical methods detect it. Limits for individual impurities are set by pharmacopoeial monographs or manufacturer specifications. How much a given impurity affects biological activity is often uncertain, and conclusions may depend on the assay used.
Serum protein binding dominates the pharmacokinetic profile. The attached chain associates strongly with albumin, shielding the peptide from enzymatic attack and slowing filtration by the kidney. This interaction extends the circulation half-life to roughly one week in humans, which supports weekly administration intervals. An oral version pairs the peptide with an absorption enhancer that transiently alters gastric epithelium, permitting limited uptake; bioavailability by that route is substantially lower than by injection.
Semaglutide belongs to the glucagon-like peptide-1 receptor agonist class, a group of synthetic peptides that imitate an incretin hormone released by intestinal L cells after food intake. Native GLP-1 circulates for only a few minutes because dipeptidyl peptidase-4 cleaves it rapidly. The hormone acts on pancreatic islets, the gastrointestinal tract, and several brain regions. Because the natural peptide is short-lived, development work concentrated on analogues that keep receptor activity while resisting enzymatic breakdown and renal clearance.
== Applications == The main advantage of paper-based microfluidic devices over traditional microfluidics devices is their potential for use in the field rather than in a laboratory. Filter paper is advantageous in a field setting because it is capable of removing contaminants from the sample and preventing them from moving down the microchannel. This means that particles will not inhibit the accuracy of paper-based assays when they are used outdoors. Paper-based microfluidic devices are also small in size (approximately a few mm to 2 cm in length and width) compared to other microfluidic platforms, such as droplet-based microfluidic devices, which often use glass slides up to 75 mm in length. Because of their small size and relatively durable material, paper-based microfluidic devices are portable. Paper-based devices are also relatively inexpensive. Filter paper is very cheap, and so are most of the patterning agents used in the fabrication of microchannels, including PDMS and wax. Most of the major paper-based fabrication methods also do not require expensive laboratory equipment. These characteristics of paper-based microfluidics make it ideal for point-of-care testing, particularly in countries that lack advanced medical diagnostic tools. Paper-based microfluidics has also been used to conduct environmental and food safety tests.

Carbamazepine, sold under the brand name Tegretol among others, is an anticonvulsant medication used in the treatment of epilepsy and neuropathic pain. It is used as an adjunctive treatment in schizophrenia along with other medications and as a second-line agent in bipolar disorder. Carbamazepine appears to work as well as phenytoin and valproate for focal and generalized seizures. It is not effective for absence or myoclonic seizures. Carbamazepine was discovered in 1953 by Swiss chemist Walter Schindler. It was first marketed in 1962. It is available as a generic medication. It is on the World Health Organization's List of Essential Medicines. In 2023, it was the 185th most commonly prescribed medication in the United States, with more than 2 million prescriptions. Photoswitchable analogues of carbamazepine have been developed to control its pharmacological activity locally and on demand using light (photopharmacology), with the purpose of reducing the adverse systemic effects of the drug. One of these light-regulated compounds (carbadiazocine, based on a bridged azobenzene or diazocine) has been shown to produce analgesia with noninvasive illumination in vivo in a rat model of neuropathic pain.
beta bend Also β-bend, beta turn or β-turn. A short segment of a polypeptide chain in which the main direction of the chain changes, i.e. a bend or turn in an otherwise linear polymeric chain. The bend is facilitated by a span of four consecutive amino acid residues where the carboxyl group of the first residue participates in hydrogen bonding with the amino group of the fourth residue.

== Legacy == Duttaroy's research focuses on how the placenta transports maternal plasma DHA and ARA to the fetus. The fetal brain development in utero is critically dependent on the maternal supply of these fatty acids in the utero. His studies have been documented through several articles. Besides, his articles have drawn many citations (Google Scholar PubMed). His other research area is cardioprotective factors in fruits and vegetables. He has discovered the anti-platelet factors from tomatoes. This discovery made by him is now widely known as Fruitflow. In 2009, Fruitflow® became the first product in Europe to obtain an approved, proprietary health claim under Article 13(5) of the European Health Claims Regulation 1924/2006 on nutrition and health claims made on foods. The EU Commission authorized the health claim "water-soluble tomato concentrate (WSTC) I and II help maintain normal platelet aggregation, which contributes to healthy blood flow." Fruitflow® is now widely available in different countries worldwide.
Kennedy was sworn in as Louisiana's junior U.S. Senator on January 3, 2017. He had resigned his position as state treasurer earlier that day. In June 2017, Kennedy "grilled" Education Secretary Betsy DeVos in a hearing before the Appropriations subcommittee on Labor, Health and Human Service, Education and Related Agencies. In the exchange, he contrasted the lack of school choice available to younger pupils in many rural areas of the country with the numerous brands of mayonnaise available at grocery stores: "Now I can go down to my overpriced Capitol Hill grocery this afternoon and choose among about six different types of mayonnaise. How come I can't do that for my kid?" Kennedy asked. The remark attracted national attention. DeVos replied that the Trump administration budget proposal would give parents and students more power and opportunity so that American education could again become "the envy of the world". Kennedy served as a volunteer substitute teacher for more than a decade in Louisiana public schools to better understand issues in the education system. He attracted comment for his manner in the Senate. A January 2018 Huffington Post article reported: "Since being elected to the Senate a year ago, Kennedy ... has made a name for himself on Capitol Hill with his wit, humor and penchant for folksy expressions―a notable feat in a place where jargon and arcane procedure tend to reign supreme". Kennedy received widespread media attention after he crossed party lines to oppose the appointment of three of Trump's U.S.
== Method == SISCAPA is an extension of the well-known gold-standard methods of stable-isotope dilution for quantitation of small molecules by mass spectrometry (MS). Rather than measure an intact protein directly by mass spectrometry, SISCAPA makes use of proteolytic digestion (e.g., with the enzyme trypsin) to cleave sample proteins into smaller peptides ideally suited to quantitation by mass spectrometry. By selecting a target peptide whose sequence occurs only in the selected target protein (a so-called “proteotypic” peptide), the target peptide can serve as a direct quantitative surrogate for the target protein (assuming the digestion process is complete, or at least reproducible). A synthetic version of the target peptide containing a stable isotope label is added in a known amount to the digested sample to serve as an internal standard (SIS). Since the target peptide and SIS are chemically indistinguishable throughout the workflow, but can be measured separately by a mass spectrometer due to the mass difference of the stable isotope label, their ratio provides the desired quantitative estimate of the target peptide amount. The SISCAPA workflow adds a specific enrichment step to the isotope dilution method in which a selected target peptide, together with its associated SIS internal standard, is captured by a sequence-specific anti-peptide antibody.

=== Skin and inflammation === ApoB lipoprotein, PCSK9, and the genes involved in cholesterol synthesis are highly expressed in the epidermis. The cutaneous expression of PCSK9 is likely important for proper skin barrier formation as ceramides, free fatty acids, and cholesterol are the three major components of the epidermal lipid barrier. Matching its function in cholesterol homeostasis, there is a gradient of PCSK9 expression in the epidermis. PCSK9 is selectively expressed in basal and spinous layer keratinocytes with little to no expression in granular layer keratinocytes. In contrast to basal layer keratinocytes, granular layer keratinocytes release large amounts of cholesterol and other lipids to form a lipid rich "mortar" in the intracellular space between keratinocytes. In addition to its likely role in epidermal lipid barrier formation, PCSK9 has also been linked to skin inflammation. For example, genetic variants of PCSK9 have been linked psoriasis, and knockdown expression of PCSK9 in keratinocytes results in increase expression of IL-36G and other keratinocyte-derived inflammatory mediators.

It is given either as a once-weekly subcutaneous injection or as an oral tablet taken once daily. The two forms use different absorption strategies, so they are not interchangeable on a milligram-for-milligram basis.
Structural modifications, including a fatty acid side chain and non-natural amino acid substitutions, slow enzymatic breakdown and promote albumin binding. These changes support once-weekly dosing rather than twice-daily administration.
The pathways involving insulin, glucagon, gastric emptying, and appetite signaling are well described. How much each pathway contributes to weight reduction in a given person is not fully established.
