=== Cell and scaffold-based cartilage regeneration === In order to restore joint cartilage after injury due to chondrocyte loss, cell therapy and chondrocyte replenishment has been shown to work in certain studies. Lying self-assembled MSCs (mesenchymal stem cells) on top of chondrocyte-laden hydrogel scaffolds has shown cell-mediated regeneration of hyaline-like cartilage. However, one drawback of this is that implantation of these scaffolds requires open-joint surgery to gather donor chondrocytes from non-weight-bearing joint cartilage areas. == Medical uses == The safety and efficacy of trastuzumab-containing combination therapies (with chemotherapy, hormone blockers, or lapatinib) for the treatment of metastatic breast cancer. The overall hazard ratios (HR) for overall survival and progression free survival were 0.82 and 0.61, respectively. It was difficult to accurately ascertain the true impact of trastuzumab on survival, as in three of the seven trials, over half of the patients in the control arm were allowed to cross-over and receive trastuzumab after their cancer began to progress. Thus, this analysis likely underestimates the true survival benefit associated with trastuzumab treatment in this population. In early-stage HER2-positive breast cancer, trastuzumab-containing regimens improved overall survival (Hazard ratio (HR) = 0.66) and disease-free survival (HR = 0.60). Increased risk of heart failure (RR = 5.11) and decline in left ventricular ejection fraction (relative risk RR = 1.83) were seen in these trials as well. Two trials involving shorter term treatment with trastuzumab did not differ in efficacy from longer trials, but produced less cardiac toxicity. The original studies of trastuzumab showed that it improved overall survival in late-stage (metastatic) HER2-positive breast cancer from 20.3 to 25.1 months. In early-stage HER2-positive breast cancer, it reduces the risk of cancer returning after surgery. The absolute reduction in the risk of cancer returning within three years was 9.5%, and the absolute reduction in the risk of death within 3 years was reduced by 3%.

=== Oceania === The white shark population is estimated to be 2,500–6,750 individuals around eastern Australia and New Zealand. In 1999, the Australian Government gave legal protection to the species under the Environmental Protection and Biodiversity Conservation (EPBC) Act, subsequently declaring it vulnerable in 2000. Similar protections exist at the state level—some predating federal law—with New South Wales, Tasmania, and Western Australia listing the species as vulnerable, and Victoria classifying it as endangered. To bolster these protections, the federal government launched the White Shark Recovery Plan in 2002 to mandate conservation research and regulate shark-related trade and tourism. An updated 2013 recovery plan reported moderate success, with 14 of its 34 original objectives completed. A study in 2012 found that Australia's white shark population is separated by the Bass Strait into genetically distinct eastern and western populations, necessitating regionalized conservation strategies. Population declines prior to protection were driven by commercial and sport fishing, along with entanglement in beach protection netting. By 2013, incidental deaths from commercial and recreational fishing had decreased, yet the population had not fully recovered. == Safety == The safety of ketosis from low-carbohydrate diets is often called into question by clinicians, researchers and the media. A common safety concern stems from the misunderstanding of the difference between physiological ketosis and pathologic ketoacidosis. There is also continued debate whether chronic ketosis is a healthy state or a stressor to be avoided. Some argue that humans evolved to avoid ketosis and should not be in ketosis long-term. The counter-argument is that there is no physiological requirement for dietary carbohydrates, as adequate energy can be made via gluconeogenesis and ketogenesis indefinitely. Alternatively, the switching between a ketotic and fed state has been proposed to have beneficial effects on metabolic and neurologic health. The effects of sustaining ketosis for up to two years are known from studies of people following a strict ketogenic diet for epilepsy or type 2 diabetes; these include short-term adverse effects leading to potential long-term ones. However, literature on longer term effects of intermittent ketosis is lacking.
=== Modern method === Deep-frozen stomachs are milled and put into an enzyme-extracting solution. The crude rennet extract is then activated by adding acid; the enzymes in the stomach are produced in an inactive form and are activated by the stomach acid. The acid is then neutralized and the rennet extract is filtered in several stages and concentrated until reaching a typical potency of about 1:15,000; meaning 1 g of extract can coagulate 15 kg of milk. One kilogram of rennet extract has about 0.7 g of active enzymes – the rest is water and salt and sometimes sodium benzoate (E211), 0.5%–1.0% for preservation. Typically, 1 kg of cheese contains about 0.3 mg of rennet enzymes.
== Arrest and trial == Pressdee was arrested on May 25, 2023. She was charged with three counts of murder and 19 counts of attempted murder. Authorities also linked her to the deaths of 17 patients. To avoid the death penalty, Pressdee pleaded guilty to all charges. She ultimately pleaded guilty to the murders of James Bartoe, 55; Gerald Shrum, 90; and Nicholas Cymbol, 43. On May 2, 2024, she was sentenced to three consecutive life terms, along with 380 to 760 additional years. She is incarcerated at the State Correctional Institution in Muncy, Pennsylvania.

== Structure == The GSTK1 enzyme is a homodimer and, like all GSTs, it contains a TRX-like domain and a helical domain. However, the GSTK1 is substantially different in its secondary structure compared to the other GSTs. The helical domain has been observed to be placed between the βαβ and ββα motifs of the TRX-like domain, rather than the TRX-like domain and the C-terminal helical domain being connected together by a short linker of alpha-helixes as normally seen in GSTs. Also, the GSTK1 dimer employs a butterfly shape and not a V-shaped crevice like in the other classes. As for the GSTK1 gene, it is ~5 kb long, has eight exons, is located on chromosome 7q34, and includes an initiator element at the transcription start site instead of a TATA or a CCAAT box.
