Large randomised trials in adults with type 2 diabetes and in adults with obesity have reported reductions in body weight and improvements in several cardiovascular risk markers. One outcome trial found a lower incidence of major adverse cardiovascular events in participants with diabetes and established cardiovascular disease. Gastrointestinal effects such as nausea and vomiting are the most frequently reported adverse events and often diminish over time. Changes in lean body mass during weight loss are an area of ongoing investigation. Effects in adolescents and in pregnancy are less well characterised, and current labelling advises against use during pregnancy.
Semaglutide is a synthetic peptide analogue of glucagon-like peptide-1, a gut hormone released after nutrient intake. The molecule contains 31 amino acid residues and differs from the native sequence at several positions. A non-natural residue at position eight resists the enzyme that normally truncates the hormone, while a lysine-linked fatty diacid side chain promotes binding to serum albumin. These two modifications extend the circulating half-life from minutes to roughly one week. The peptide is produced by solid-phase synthesis followed by selective acylation, and its identity and purity are confirmed by spectrometric and chromatographic techniques.
Identity and purity are assessed with reversed-phase high-performance liquid chromatography, which separates the peptide from related impurities by hydrophobicity. Mass spectrometry confirms molecular weight and detects truncation or modification products. Peptide mapping after enzymatic digestion verifies the amino acid sequence. Quantitation is often performed by LC-MS/MS or by immunoassay, and the two approaches can give different values because they measure different things. Method validation parameters such as accuracy, precision, and limit of quantitation are reported alongside results.
Certificate of analysis documents from suppliers typically report purity by chromatographic area, water content, and counter-ion identity. Independent verification is advisable because reported values can be generated under differing conditions. Impurity profiles matter for research use, where aggregates, deamidation products, and residual solvents may influence experimental results. Container, lot, and chain-of-custody records support traceability. Analytical results are method-dependent, so comparisons between laboratories require the same procedure and reference standards.
Lyophilized peptide material is typically stored at or below -20 °C, with -80 °C used for longer-term archives. Vials should remain sealed and desiccated because moisture promotes aggregation and hydrolysis. Repeated freeze-thaw cycles are avoided since they can alter peptide conformation and reduce recovery. Once reconstituted, solutions are generally kept at 2-8 °C and used within a defined window. Stability beyond those windows depends on buffer composition and concentration, and exact limits are product-specific rather than universal.

Semaglutide is a synthetic peptide that acts as an agonist at the glucagon-like peptide-1 receptor. It is a structural analogue of human GLP-1(7-37), modified to resist enzymatic degradation by dipeptidyl peptidase-4. The peptide backbone contains alpha-aminoisobutyric acid at position 8, a substitution that stabilises the helix and slows cleavage. A fatty diacid side chain attached through a linker at lysine 34 promotes binding to serum albumin, which extends the circulating half-life. These two modifications together allow less frequent administration than native GLP-1 requires.
Activation of the GLP-1 receptor couples to Gs signalling and raises intracellular cyclic AMP in pancreatic beta cells. The resulting insulin release depends on prevailing glucose concentrations, so the effect is greater when glucose is elevated. Receptor engagement also suppresses glucagon secretion and slows gastric emptying, which flattens post-meal glucose excursions. In the central nervous system, signalling in hypothalamic and brainstem regions is associated with reduced appetite and lower energy intake. Studies continue to examine effects on cardiac, renal and hepatic endpoints; whether those benefits are independent of weight change remains an open question.
The compound binds the GLP-1 receptor on pancreatic beta cells and other tissues, activating a G-protein signaling cascade that raises intracellular cyclic AMP. This action increases glucose-dependent insulin secretion when blood glucose is elevated, while binding also slows gastric emptying and reduces glucagon release. In the central nervous system, receptor activation in the hypothalamus and brainstem contributes to reduced appetite. The fatty acid chain binds albumin, which protects the peptide from renal filtration and enzymatic degradation. This albumin binding is central to its extended circulation time.
Native GLP-1 is degraded rapidly by dipeptidyl peptidase-4. Semaglutide resists this cleavage because alanine at position 8 is replaced by alpha-aminoisobutyric acid. A second substitution at position 34 introduces arginine, which further stabilizes the peptide. The most distinctive modification is a spacer and C18 fatty diacid attached at lysine 26, enabling strong albumin affinity. These three changes together produce a half-life measured in days rather than minutes, and the same structural logic underlies other long-acting analogs in this class.
Semaglutide is a synthetic peptide analog of glucagon-like peptide-1, a hormone released from intestinal L-cells after food intake. It contains 31 amino acids and differs from native GLP-1 through modifications that slow enzymatic breakdown. The peptide was developed to extend the short circulating half-life of endogenous GLP-1, which is measured in minutes. Researchers introduced the compound in the early 2010s. Two backbone changes and a fatty acid side chain define its structure, distinguishing it from earlier GLP-1 receptor agonists.
Reported molecular weight is approximately 4113.6 daltons for the free base, and the peptide is supplied as a lyophilized powder or in buffered liquid form depending on the intended use. It is freely soluble in water when formulated with appropriate excipients, though the unconjugated peptide shows limited stability at neutral pH over long periods. Analytical characterization typically relies on reversed-phase high-performance liquid chromatography and mass spectrometry. Purity specifications for research-grade material commonly exceed ninety-five percent by area. Isotopic and impurity profiles differ between suppliers.
Two principal therapeutic variants exist under separate regulatory filings, one indicated for glycemic control in type 2 diabetes and one for chronic weight management. Both use the same active molecule; differences lie in formulation strength, titration schedule, and labeling. Regulatory agencies in the United States and European Union approved injectable forms in 2017 and 2018 respectively. An oral tablet formulation received approval later, using a carrier molecule to enhance absorption across the gastric epithelium. Labeling differs by jurisdiction and by indication.
The distinction between established facts and open questions matters here. That the peptide binds the GLP-1 receptor and stimulates insulin release in a glucose-dependent manner is well documented. How individual variability in receptor density, gastric emptying rate, and gut microbiome composition shapes response remains an active research area. Long-term outcomes beyond five years of continuous use are not yet fully characterized in published trials, and several extension studies are ongoing.
32. ArXiv [Preprint]. 2026 Sep 17:arXiv:2609.05858v2. Fluidization in Growth-Induced Morphogenesis. Wu M. Elastic buckling has explained shape formation in growing tissues, yet the role of tissue fluidity remains elusive. We derive a minimal fluidized growth-elasticity model as a nonlinear analogue of Maxwell rheology. Analysis of a growing strip reveals a different picture of growth-induced morphogenesis: rather than emerging at a critical stress, symmetry breaking develops continuously during growth. Fluidity regulates stress evolution, the rate of shape-symmetry breaking, and flow patterns, establishing it as an active regulator of morphogenesis beyond its intuitive role in stress relaxation. Curium is a synthetic chemical element; it has symbol Cm and atomic number 96. This transuranic actinide element was named after eminent scientists Marie and Pierre Curie, both known for their research on radioactivity. Curium was first intentionally made by the team of Glenn T. Seaborg, Ralph A. James, and Albert Ghiorso in 1944, using the cyclotron at Berkeley. They bombarded the newly discovered element plutonium (the isotope 239Pu) with alpha particles. This was then sent to the Metallurgical Laboratory at the University of Chicago where a tiny sample of curium was eventually separated and identified. The discovery was kept secret until after the end of World War II. The news was released to the public in November 1947. Most curium is produced by bombarding uranium or plutonium with neutrons in nuclear reactors – one tonne of spent nuclear fuel contains ~20 grams of curium. Curium is a hard, dense, silvery metal with a high melting and boiling point for an actinide. It is paramagnetic at ambient conditions, but becomes antiferromagnetic upon cooling, and other magnetic transitions are also seen in many curium compounds. In compounds, curium usually has valence +3 and sometimes +4; the +3 valence is predominant in solutions. Curium readily oxidizes, and its oxides are a dominant form of this element. It forms strongly fluorescent complexes with various organic compounds. If it gets into the human body, curium accumulates in bones, lungs, and liver, where it promotes cancer.
Despite criticism and international opposition to his controversial anti-narcotics drive, Duterte's domestic approval rating remained relatively high throughout his presidency. Duterte began his presidency with a high trust rating of 79% and 91% (the highest of any Philippine official since 1999), from pollsters Social Weather Stations (SWS) and Pulse Asia, respectively. His net satisfaction rating reached its lowest at 45% in July 2018, attributed to inflation during the period as well as Duterte calling God "stupid" for the concept of original sin in the Bible; eventually, his satisfaction rating rose to 60% by the end of the year. Halfway through his term, Duterte's approval rating reached 79%, the highest among his predecessors at this stage in their presidencies. It rose to 87% by the end of 2019 due to the successful hosting of the 2019 Southeast Asian Games as well as the administration's poverty reduction programs. Amid the COVID-19 pandemic, Pulse Asia reported in September 2020 that 84% of Filipinos approved of the administration's efforts to contain the virus and provide assistance to those who lost their jobs; 92% of the poll respondents also agreed Duterte had "done well" in preventing the contagion in the country. Duterte ended 2021 with an approval rating of 60%, wherein higher net satisfaction was observed among those who have been or are willing to get vaccinated. Duterte maintained his high approval and trust ratings as he approached the end of his term.
At half-neutralization the ratio [A−]/[HA] = 1; since log(1) = 0, the pH at half-neutralization is numerically equal to pKa. Conversely, when pH = pKa, the concentration of HA is equal to the concentration of A−. The buffer region extends over the approximate range pKa ± 2. Buffering is weak outside the range pKa ± 1. At pH ≤ pKa − 2 the substance is said to be fully protonated and at pH ≥ pKa + 2 it is fully dissociated (deprotonated). If the pH is known, the ratio may be calculated. This ratio is independent of the analytical concentration of the acid. In water, measurable pKa values range from about −2 for a strong acid to about 12 for a very weak acid (or strong base). A buffer solution of a desired pH can be prepared as a mixture of a weak acid and its conjugate base. In practice, the mixture can be created by dissolving the acid in water, and adding the requisite amount of strong acid or base. When the pKa and analytical concentration of the acid are known, the extent of dissociation and pH of a solution of a monoprotic acid can be easily calculated using an ICE table.

Animal studies suggest that RB-101 is also likely to be useful in relieving the symptoms of acute opioid withdrawal and in the management of opioid dependence. A significant advantage of inhibiting the breakdown of endogenous opioid peptides rather than stimulating opioid receptors with exogenous drugs is that the levels of opioid peptides are only increased slightly from natural levels, thus avoiding overstimulation and downregulation of the opioid receptors. This means that even when RB-101 is used in high doses for extended periods of time, there is no development of dependence on the drug or tolerance to its analgesic effects. Consequently, even though RB-101 is able to produce potent analgesic effects via the opioid system, it is unlikely to be addictive. Unlike conventional opioid agonists, RB-101 also failed to produce respiratory depression, which suggests it might be a much safer drug than traditional opioid painkillers. RB-101 also powerfully potentiated the effects of traditional analgesics such as ibuprofen and morphine, suggesting that it could be used to boost the action of a low dose of normal opioids which would otherwise be ineffective. RB-101 itself is not orally active and so has not been developed for medical use in humans, however modification of the drug has led to newer orally acting compounds such as RB-120 and RB-3007, which may be more likely to be adopted for medical use if clinical trials are successful.
Aryl nitriles can be synthesized via the Sandmeyer reaction of diazonium salts with copper(I) cyanide or by the Rosenmund-von Braun reaction (direct reaction of an aryl bromide with copper(I) cyanide). Conversion of thiocyanate with aromatic carboxylic acids, known as Letts nitrile synthesis, can be carried out using potassium thiocyanate; lead thiocyanate generally provides higher yields. Aryl iodides can be converted into aromatic nitriles under palladium catalysis with trimethylsilyl cyanide. For example, iodobenzene reacts with trimethylsilyl cyanide in the presence of tetrakis(triphenylphosphine)palladium(0) (Pd(PPh3)4) to form benzonitrile. Another palladium-catalyzed route (also employing Pd(PPh3)4) is the decarbonylation of aromatic acyl cyanides. Palladium-catalyzed cyanation of aryl chlorides with potassium cyanide or potassium hexacyanidoferrate(II) has likewise been reported. Quinones can react with trimethylsilyl cyanide to give silylated cyanohydrins, which are subsequently aromatized using phosphorus tribromide. A further approach involves reaction of aryl Grignard or aryllithium reagents with dimethylmalonitrile. Aromatic nitriles are often prepared in the laboratory from the aniline via diazonium compounds. This is the Sandmeyer reaction. It requires transition metal cyanides.

==== Cytotoxicity inhibition ==== One way that honokiol acts as a neuroprotective is through cellular regulation and subsequent inhibition of cytotoxicity. Two mechanisms used to achieve this inhibition are GABAA Modulation and Ca2+ Inhibition. Cytotoxicity inhibition may be the neuroprotective mechanism of honokiol. Honokiol has also been shown to inhibit repetitive firing by blocking glutamate.
After the acquisition by Disney in 2009, Marvel films began to be marketed at the Innoventions attraction in Tomorrowland at Disneyland. For Iron Man 3, the exhibit, entitled "Iron Man Tech Presented by Stark Industries", featured the same armor display that was shown at the 2012 San Diego Comic-Con, with the Marks I-VII and the new Mark XLII. In addition, there was a simulator game, titled "Become Iron Man", that used Kinect-like technology to allow the viewer to be encased in an animated Mark XLII armor and take part in a series of "tests", in which you fire repulsor rays and fly through Tony Stark's workshop. The game was guided by J.A.R.V.I.S., who is voiced again by Paul Bettany. The exhibit also had smaller displays that included helmets and chest pieces from the earlier films and the gauntlet and boot from an action sequence in Iron Man 3. The exhibit for Thor: The Dark World was called "Thor: Treasures of Asgard", and featured displays of Asgardian relics and transports guests to Odin's throne room, where Thor greets them. Captain America: The Winter Soldier's exhibit, "Captain America: The Living Legend and Symbol of Courage", featured a meet and greet experience. From May to September 2017, Disneyland Resort featured the "Summer of Heroes", which saw members of the Guardians and Avengers appear throughout the Disneyland Resort.
== Precautions == Californium that bioaccumulates in skeletal tissue releases radiation that disrupts the body's ability to form red blood cells. The element plays no natural biological role in any organism due to its intense radioactivity and low concentration in the environment. Californium can enter the body from ingesting contaminated food or drinks or by breathing air with suspended particles of the element. Once in the body, only 0.05% of the californium will reach the bloodstream. About 65% of that californium will be deposited in the skeleton, 25% in the liver, and the rest in other organs, or excreted, mainly in urine. Half of the californium deposited in the skeleton and liver are gone in 50 and 20 years, respectively. Californium in the skeleton adheres to bone surfaces before slowly migrating throughout the bone. The element is most dangerous if taken into the body. In addition, californium-249 and californium-251 can cause tissue damage externally, through gamma ray emission. Ionizing radiation emitted by californium on bone and in the liver can cause cancer.
=== Collateral consequences === Some authors have suggested that the collateral consequences of criminal conviction are more serious than the legal penalties. In many cases, statutes do not require that convicts are informed of these consequences. Many felons cannot be employed by the federal government or work in government jobs, as they do not meet the standards to gain security clearance. Felons convicted of distributing or selling drugs may not enlist in the military. Certain states are financially incentivized to exclude criminals from access to public housing. All states receive less federal highway funding if they fail to revoke or suspend driver's licenses of drug-related felons. Collateral consequences, and felon disenfranchisement in particular, have historically been at least partially racially motivated.

Native GLP-1 is degraded within minutes by dipeptidyl peptidase-4 and neutral endopeptidases. Semaglutide carries a non-natural amino acid at position eight that blocks that cleavage, and a fatty diacid side chain that binds albumin. The result is a much longer duration of action than the native hormone.
The fatty diacid chain associates strongly with serum albumin, which keeps the peptide in circulation and shields it from rapid renal clearance. Bound peptide is released gradually, producing a sustained receptor signal. This mechanism also reduces the peak-to-trough variation seen with shorter-acting analogues.
The balance between peripheral receptor activation and signalling in the central nervous system is not fully resolved. The extent to which slowed gastric emptying accounts for reduced energy intake, compared with direct effects on appetite circuits, is debated. Long-term effects on lean mass and on tissues outside the gut and brain are still under study.
The lyophilized powder is dissolved in a suitable solvent, often sterile water or a buffered diluent, with gentle mixing rather than vigorous shaking. Foaming and shear should be avoided because they can promote aggregation. The resulting solution is then stored cold and protected from light.
